Mechanism of Action
Its mechanism of action is expressed through the inhibition of the adhesion of the surface protein FimH. By directly binding FimH to D-mannose, which is linked to a carrier protein responsible for receptor specificity.
The first stage of adhesion involves the mannose-sensitive binding of FimH to the epithelium. The wall of the urinary bladder is coated with various proteins glycosylated with mannose, such as the Tamm-Horsfall protein (THP), which directly interferes with the adhesion of microbes to the mucosa. The Tamm-Horsfall protein (THP) can bind to E. Coli with a specific bond, which can be inhibited by exogenous D-mannose.
By inhibiting the adhesion of microbes to the epithelium of the urinary tract, D-mannose mimics the function of the epithelial barrier of the urinary tract.
